T-Vector pMD20 is a linearized vector with a single 3’-terminal thymidine at both ends. The T-overhang ends at the cloning site improve the efficiency of ligation of PCR products that contain A-overhangs at 3’-ends. T-Vector pMD20 is derived from pUC19 backbone and made some modifications in the coding sequence which can reduce the false positive rate and improve the positive colonies screened by blue/white color on ampicillin/IPTG/X-Gal LB plates. In addition, SP6 promoter has been inserted in the upstream of multiple cloning site. Therefore, the inserted DNA fragments can be transcribed by SP6 RNA Polymerase (Cat. #2520). T-Vector pMD20 is supplied with a Control Insert (500 bp) DNA for a positive control reaction.
